In vivo localization of injected carcinogen by fluorescence quartz rod microscopy.
نویسنده
چکیده
A technic whereby carcinogenic hydrocarbon particles injected into living animals can be identified by fluorescence microscopy of living lung fields is described. The meth od utilizes high pressure mercury light sources, with appropriate filters at the source and microscope optics for fluorescence examination; a quartz rod as light carrier to the lung field; and special thoracotomy on living rats and rabbits for exposure of the area to be examined. The particles stood out wherever localized as long as 5 months after injec tion. They could be studied by display on a television camera and photography of the monitor image. Film was too slow for in vivo instant recording. Damage, as revealed by ordinary microscopy, was not very obvious, and further in vivo methodology to detect alterations in the physicochemical nature of areas involved is suggested. Liver, spleen, kidney, brain, muscle, bone marrow, and skin also revealed the particles after post mortem fresh examination.

Comparative Studies of High Contrast Fluorescence Imaging Efficiency of Silica-coated CdSe Quantum Dots with Green and Red Emission
Herein we report the possibility of using green and red emitting silica-coated cadmium selenide (CdSe) quantum dots (QDs) for remarkable stem and cancer cellular imaging, efficient cellular uptake and fluorescence imaging of semi and ultra-thin sections of tumor for in vivo tumor targeted imaging applications. The comparative studies of high contrast cellular imaging behaviours of the silica-co...
متن کاملCofilin nuclear-cytoplasmic shuttling affects cofilin-actin rod formation during stress.
Cofilin protein is involved in regulating the actin cytoskeleton during typical steady state conditions, as well as during cell stress conditions where cofilin saturates F-actin, forming cofilin-actin rods. Cofilin can enter the nucleus through an active nuclear localization signal (NLS), accumulating in nuclear actin rods during stress. Here, we characterize the active nuclear export of cofili...
متن کاملUsing a quartz paraboloid for versatile wide-field TIR microscopy with sub-nanometer localization accuracy.
Illumination based on objective-type total internal reflection (TIR) is nowadays widely used in high-performance fluorescence microscopy. However, the desirable application of such setups for dark-field imaging of scattering entities is cumbersome due to the spatial overlap of illumination and detection light, which cannot be separated spectrally. Here, we report a novel TIR approach based on a...
متن کاملStudying Biomolecule Localization by Engineering Bacterial Cell Wall Curvature
In this article we describe two techniques for exploring the relationship between bacterial cell shape and the intracellular organization of proteins. First, we created microchannels in a layer of agarose to reshape live bacterial cells and predictably control their mean cell wall curvature, and quantified the influence of curvature on the localization and distribution of proteins in vivo. Seco...
متن کاملFibered Confocal Fluorescence Microscopy for the Noninvasive Imaging of Langerhans Cells in Macaques
Purpose We developed a new approach to visualize skin Langerhans cells by in vivo fluorescence imaging in nonhuman primates. Procedures Macaques were intradermally injected with a monoclonal, fluorescently labeled antibody against HLA-DR molecule and were imaged for up to 5 days by fibered confocal microscopy (FCFM). Results The network of skin Langerhans cells was visualized by in vivo fib...
